چکیده
BACKGROUND 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) has a long half-life of 5-10 years in human beings as a result of its high lipophilicity, and little or no metabolism. We monitored TCDD, its form, distribution, and elimination in Victor Yushchenko after he presented with severe poisoning. METHODS In late December, 2004, a patient presented with TCDD poisoning; the levels in his blood serum (108000 pg/g lipid weight) were more than 50 000-fold greater than those in the general population. We identified TCDD and its metabolites, and monitored their levels for 3 years using gas chromatography and high-resolution mass spectrometry in samples of blood serum, adipose tissue, faeces, skin, urine, and sweat, after they were extracted and cleaned with different organic solvents. FINDINGS The amount of unmodified TCDD in the samples that were analysed accounted for about 60% of TCDD eliminated from the body during the same period. Two TCDD metabolites-2,3,7-trichloro-8-hydroxydibenzo-p-dioxin and 1,3,7,8-tetrachloro-2-hydroxydibenzo-p-dioxin-were identified in the faeces, blood serum, and urine. The faeces contained the highest concentration of TCDD metabolites, and were the main route of elimination. Altogether, the different routes of elimination of TCDD and its metabolites accounted for 98% of the loss of the toxin from the body. The half-life of TCDD in our patient was 15.4 months. INTERPRETATION This case of poisoning with TCDD suggests that the design of methods for routine assessment of TCDD metabolites in human beings should be a main aim of TCDD research in the metabolomic era. TEMPORAL TRENDS IN ETHOXYRESORUFIN-O-DEETHYLASE ACTIVITY OF BROOK TROUT (SALVELINUS FONTINALIS) FED 2,3,7,8-TETRACHLORODIBENZO-p-DIOXIN
Changes in ethoxyresorufin-O-deethylase (EROD) activity were monitored through an extended 6-month dietary exposure to determine the relationship between EROD activity and uptake of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) in brook trout, Salvelinus fontinalis. Brook trout were fed labeled TCDD during a 4-week loading phase and an 11-week maintenance phase to achieve whole-body concentrations...
متن کاملSynthesis of new steric and electronic analogs of 2,3,7,8-tetrachlorodibenzo-p-dioxin.
In contrast to the determination of a chemical reaction mechanism, the molecular mechanism for xenobiotic toxicity among polychlorinated dibenzo-p-dioxins is a problem of imposing complexity. These compounds are often lethal at such low dosages that identification of in vivo localization sites, or of metabolites, is especially difficult. Even though recent work (1, 2) has identified specific en...
متن کاملEffects of Perinatal 2,3,7,8-Tetrachlorodibenzo-P-Dioxin Exposure on Development of Taste Preference in Rat Offspring
2,3,7,8-Tetrachlorodibenzo-P-Dioxin (TCDD) is a highly toxic man-made chemical found ubiquitously in the environment. Maternal exposure to TCDD during pregnancy has been reported to induce deficits in higher brain functions of off-spring, such as working memory in a radial arm maze, operant responding in running wheels or two-lever chamber, and discrimination reversal learning [1-4]. We also re...
متن کامل2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) reduces Leishmania major burdens in C57BL/6 mice.
Acute exposure to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) can suppress adaptive immunity. In this study, pre-exposure of Leishmania major-infected mice to TCDD caused a dose-dependent and unexpected decrease in parasite burdens on day 20 after infection. In contrast, TCDD-mediated lymphoid atrophy, suppressed antibody levels, and enhanced interleukin-2 production were observed as expected. T...
متن کاملEffects of dietary exposure of mink (Mustela vison) to 2,3,7,8-tetrachlorodibenzo-p-dioxin, 2,3,4,7,8-pentachlorodibenzofuran, and 2,3,7,8-tetrachlorodibenzofuran on reproduction and offspring viability and growth.
This study assessed the effects of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), 2,3,4,7,8-pentachlorodibenzofuran (PeCDF), and 2,3,7,8-tetrachlorodibenzofuran (TCDF) on the reproductive performance of female mink (Mustela vison) and the viability and growth of their offspring. Nine adult female mink were randomly assigned to one of 13 dietary treatments (one control and four doses each of TCDD, ...
